The Blade character has a long history in the Marvel universe, but wasn't popular until the movie dropped in 1998.
Deacon Frost is in the comics, and pretty close to how he was in the movie.
Whistler is not from the main 616 universe. But, fun fact! The Blade movies exist in their own universe! (26320)
Whistler is based on a character named Jamal Afari.
David S Goyer wrote the story for this and a ton, a ton of other comic movies. Most people don't know how much he's had his hand in comic movies. But, once you get a sense of his style, you can see it. And, it's not always great. Blade 1 and 2, I believe, are his best work. He also directed Blade Trilogy and Jay thinks it was absolutely terrible.
Stephen Norrington directed Blade and it's a miracle that it was good. (he's not a great director) Blade II was directed by Guillermo Del Toro and you can see the difference in style, design and fight scenes. Jay says that Blade II is the rare sequel that is better than the original. 
We don't know much about the new Blade movie that they are making. It starts filming this July and Mahershala Ali is the lead role, but that's it. We know he spoke in the post credits scene for Eternals. Big questions: Will it be PG 13 or rated R?  The current Marvel movies haven't done anything "dark" yet, but there is plenty of material for them to pull from.
